Introduction Semi-Synthesized/ Synthesized Antibody Libraries

Semi-synthetic antibody library is created by combining naturally occurring antibody libraries with synthetic diversity. It usually involves synthesizing DNA oligonucleotides to produce a set of different complementary determining regions (CDRs), which are then combined with a constant antibody framework derived from natural sources such as human or animal B cells. The synthesized CDR introduces diversity into the library, resulting in antibodies targeting a wide range of epitopes. The semi-synthetic antibody library provides a compromise between the natural diversity of the immune system and the controlled diversity achieved through synthetic methods.

Synthesized antibody library, also known as a De novo library, is a synthetic method that utilizes techniques such as DNA synthesis or phage display to design and synthesize complete antibody variable regions, including framework regions and CDRs, without relying on naïve antibody libraries.

The distinction between naïve and synthetic antibody libraries depends on the source of immunoglobulin genes. Meanwhile, the difference between a semi-synthesized and synthesized antibody library lies in that the former consists of part of naïve antibody segments, such as light chain or heavy chain, and only one of them is synthesized in vitro; while the synthesized one is totally derived from an artificial synthesis by PCR in vitro.

Design
The design phase involves selecting antibody frameworks and complementarity-determining regions (CDRs) based on existing antibody sequences or structural data. Synthetic CDR antibodies can also be designed to introduce specific functionalities or enhance binding properties.

Synthesis
Synthetic DNA encoding the designed antibody sequences, including both framework regions and CDRs, is synthesized using chemical or enzymatic methods.

Assembly
The synthesized DNA fragments are assembled into antibody expression vectors using techniques such as PCR, ligation, or Gibson assembly. These vectors can then be introduced into expression systems such as bacterial, yeast, or mammalian cells for antibody production.

Screening and Selection
The constructed antibody libraries are screened and selected for antibodies with desired properties using high-throughput screening methods. This may involve techniques such as phage display, yeast display, or ribosome display, depending on the library format and application.
