The immune system of camelids (llamas, alpacas and alpacas) is different from that of other animals in that their serum contains, in addition to the traditional IgG, an antibody to VHH that has lost its light chain.VHH is only 13-15 kDa in size, and is characterized by its small size, good stability, high solubility, and ability to be cleared from the bloodstream and penetrate deep into the tissues quickly, and it has a unique antigen-binding property compared to antibody fragments. VHH has been widely used in the field of antigen binding. Utilizing these advantages, VHH is widely used to become a biosensor, affinity capture, protein crystallization, molecular imaging, diagnosis and treatment of diseases.

Alpha Lifetech provides personalized phage antibody library construction, screening, expression and antibody purification strategies to meet the different needs of our customers. We can use various types of antigens to immunize alpacas and construct VHH immune libraries based on phage display technology according to the needs of our customers. Our VHH immune libraries are diversified and can effectively produce VHH with high affinity to the original antigen, and we keep pre-made natural immune libraries in our laboratory, which do not require immunization of animals and greatly shorten the preparation time of VHH. We can also construct VHH-based synthetic libraries and semi-synthetic libraries according to customer needs. We isolate peripheral blood mononuclear cells (PBMC) from alpacas, and clone the acquired VHH gene into phage display vectors, which can generate a huge library of VHH antibodies through reverse transcription and polymerase chain reaction. We utilize the antigens required by our customers, and this library undergoes multiple rounds of screening and enrichment to obtain VHH antibodies that meet the needs of our customers for research, diagnosis or therapy.

Alpha Lifetech has a comprehensive recombinant antibody expression platform that can be utilized for VHH expression using different protein expression systems according to customer needs. Mammalian expression systems are the first choice for protein expression in humans or other mammals and are most commonly used in scientific research. Mammalian expression systems can correctly and efficiently recognize the synthesis, processing and secretion signals of eukaryotic proteins, have structure and function similar to natural proteins, can be modified post-translationally such as glycosylation, phosphorylation and acetylation, and have simple expression and purification processes, making it easy for transfection of recombinant plasmids. However, mammalian expression systems for protein passages usually have the disadvantages of low yield and long preparation time. Alpha Lifetech has been focusing on the optimization and research of mammalian expression systems for many years, and can provide customers with high yield and good quality VHH expression services. We are good at transient and stable expression of VHH using HEK293 or CHO cell lines for protein.

We can also prepare soluble proteins using the E. coli prokaryotic expression system, which optimizes protein expression conditions to avoid inclusion body expression of proteins and ensure protein activity. Alpha Lifetech also has a proven baculovirus/insect cell protein expression system for the production and expression of viral recombinant proteins from baculovirus-infected insect cells. This expression system facilitates protein solubility and post-translational modifications. Also, the baculovirus/insect cell expression system allows for the simultaneous expression of multiple genes and can produce higher yields of intracellular and secreted proteins. Our protein expression platform can be used for VHH small-scale and large-scale production to meet the different needs of our customers.
