VHH Antibody Library Construction Service

Introduction Of VHH Antibody Library Construction Service

VHH antibodies are derived from camelids ( camelids, llamas, alpacas) and sharks, and contain only heavy chain antibodies, which are the smallest binding-capable antibody fragments available, with a size of only 12-15 kDa. camelids have a distinctive immune system, and in addition to possessing the prototypical antibody, their sera contain an antibody that has lost its light chain, which makes them an ideal animal source to obtain the VHH antibodies.VHH is small in size and have good water solubility, stability, acid resistance, low immunogenicity and unique antigenic epitopes that are rapidly cleared from the bloodstream and penetrate deep into tissues. These unique properties make them widely used in biosensing, affinity capture, molecular imaging, diagnostics and therapeutics of diseases.VHHs are usually derived from immune or natural libraries and displayed and screened by phage or yeast libraries. Among them, phage display technology is most commonly used, and specific VHH antibodies can be produced by appropriate screening strategies. In addition, VHH is easy to produce in E. coli on a large scale and has unique antigen-binding properties compared to other recombinant antibodies.

What We Can Offer?

Alpha Lifetech is dedicated to providing VHH library construction services for various antigens, and can deliver VHH phage display libraries with a library capacity of 1 × 10^9 by utilizing the advantages of our antibody discovery platform. We are good at providing a variety of VHH antibody library construction services, such as natural, immune, and synthetic libraries, according to the needs of our customers, and based on our many years of experience in recombinant antibody library construction and screening, in order to screen the ideal VHH antibody for our customers. Our phage platform has a variety of phages such as M13, T4, T7 and λ to meet the different needs of customers. The VHH libraries we constructed have the advantages of large library capacity and rich diversity, and can produce high-affinity VHH antibodies. We provide personalized library construction services according to customer needs. We provide a wide range of phage vectors, including PMECS, pComb3X and pCANTAB 5E, and modify them according to customer needs. We can provide a variety of E. coli host bacteria such as TG1 E. coli, XL1-Blue and ER2738, which can be used for phage infection.

Alpha Lifetech has advanced technology in the field of recombinant antibody construction and screening, and constantly optimizes the ability to construct antibody libraries. We not only have natural antibody libraries, but also specialize in the construction of VHH immune libraries, which is beneficial to the generation of more diversified and high affinity antibodies. We immunize the prepared antigens against camelids such as alpacas, and test the antibody potency by ELISA so that the antibody titers reach the desired level. The peripheral blood of the animals was drawn to obtain PBMC and total RNA was extracted, and the VHH gene was cloned into a phage display vector by reverse transcription and polymerase chain reaction. By electrotransformation, we could prepare a huge library of VHH antibodies. We screen and enrich the library to produce VHH antibodies for research, diagnostic or therapeutic use to the satisfaction of our customers. We also have many years of experience in recombinant antibody expression and purification. Prior to the delivery of the antibody, the prepared VHH antibody will be subjected to rigorous characterization and functional identification, including Western Blot, ELISA, immunoprecipitation, flow cytometry or immunohistochemistry.
