VHH antibody is derived from camelids (e.g., dromedary camel, camel, alpaca, and alpaca), and contains only one variable region of heavy chain, lacking the CH structural domains of the light and heavy chains of the traditional Fab fragment.VHH has a very small molecular weight of only 15 KDa, which is much smaller than that of the full-length antibody (150-160 kDa) or other antibody fragments (Fab ~ 50 kDa, scFv ~ 25 kDa). With the advantages of high solubility, stable structure, easy mass production, selective binding to specific antigens and strong ability to penetrate the blood-brain barrier, VHH is often used in academic research, immunodiagnostics, therapeutic antibody development, cell therapy and environmental testing, etc., and has a wide range of applications.VHH has a great potential to improve drug bioavailability, development of bispecific antibodies, and humanization of antibodies, and can be expressed by both eukaryotic and prokaryotic systems.

Mammalian cell expression systems can post-translationally modify and appropriately fold proteins. However, under general conditions, mammalian expression systems have the disadvantages of insufficient antibody expression yields, long preparation periods and labor-intensive. In mammalian cell protein expression, Alpha Lifetech has a comprehensive and mature platform to optimize various expression conditions and reagents to achieve high yields of protein expression in a shorter period of time. We meet the different needs of our customers and can realize the preparation of VHH antibodies from small scale to large scale. We have mammalian cell lines such as HEK293 and CHO for transient expression and stable cell line development. Our mammalian cells can be cultured in suspension culture or serum-free suspension cell high-density culture, enabling large-scale preparation of antibodies. We can prepare VHH antibodies expressed by CHO cells, and the speed of transient antibody expression in CHO cells saves our customers a lot of time. Our expression platform can prepare VHH antibodies with low endotoxin levels within 4 weeks.

The E. coli expression system is also the most commonly used method for the production of recombinant antibodies due to its advantages of fast growth, ease of handling, high protein yield and low cost. Alpha Lifetech can provide customers with suitable strains to express VHH antibodies using E. coli in the form of secretory expression in the periplasm, soluble expression in the cytoplasm and insoluble expression in the form of inclusion bodies. However, the protein in inclusion body form is usually inactive, which is not conducive to VHH expression and purification. Alpha Lifetech can perform a series of optimization of conditions such as induction temperature, induction time and IPTG concentration for VHH antibody expression, so that a large number of soluble VHH antibodies can be expressed and purified. We also fused some soluble tags to the N-terminus or C-terminus, thus enhancing the expression of functional VHH in the cytoplasm. Yeast cells have both the advantages of E. coli protein expression (fast growth, genetic simplicity, high yield, low cost) and the characteristics of eukaryotic systems (post-translational modification, secretory expression), which make them ideally suited for large-scale antibody production. The yeast expression system developed by Alpha Lifetech can help customers prepare high quality VHH antibody with high expression amount.
In order to meet the needs of our customers, we will provide codon professional optimization tools free of charge to help customers optimize the antibody gene sequences, so as to achieve soluble expression of VHH antibody in mammalian expression system. We will provide various types of affinity tags such as His, Flag, GST, Myc, SUMO and HA according to customer's needs, and purify VHH antibody by affinity chromatography such as cation exchange chromatography, IMAC and SEC. We will use SUMO protease to cleave the C-terminus of SUMO to remove the purified VHH antibody tag. We can also purify the VHH antibody using protein A and G agarose columns. After purification of VHH, we will use immunofluorescence, immunoblotting, immunoprecipitation and other methods to verify the VHH, so as to ensure the quality of antibody purification.
